Mechanisms for the maintenance and eventual degradation of neurofilament proteins in the distal segments of severed goldfish Mauthner axons
Cellular mechanisms that might affect the degradation of neurofilament proteins (NFPs) were examined in the distal segments
of severed goldfish Mauthner axons (M-axons), which do
not degenerate for more than 2 months after severance. Calpain
levels, as determined by reactivity to a polyclonal antibody,
remained constant for 80 d postseverance in distal segments
of M-axons and then declined from 80 to 85 d
postseverance. Calpain activity in rat brain, as determined by a
spectrophotometric assay, was much higher than calpain activity
in control and severed goldfish brain, spinal cord, muscle,
or M-axons. Calpain activity was extremely low in M-axons
compared with that in all other tissues and remained low for up
to 80 d postseverance in distal segments of M-axons. Phosphorylated
NFPs, as determined by Stains-All treatment of SDS
gels, were maintained for up to 72 d postseverance and then decreased noticeably at 75 d postseverance when NFP breakdown
products appeared on silver-stained gels. By 85 d postseverance,
phosphorylated NFPs no longer were detected, and
NFP breakdown products were the most prominent bands on
silver-stained gels. These results suggest that the distal segments
of M-axons survive for months after severance, because
NFPs are maintained in a phosphorylated state that stabilizes
and protects NFPs from degradation by low levels of calpain
activity in the M-axon; the distal segments of severed M-axons
degenerate eventually when NFPs no longer are maintained in
a phosphorylated state and become susceptible to degradation,

Hardware Accelerating the Optimization of Transaction Schedules via Quantum Annealing by Avoiding Blocking
The isolation property of database theory guarantees to avoid problems of not synchronized parallel execution of several transactions. In this paper we propose an algorithm for an optimal transaction schedule for the different cores of a multi-core CPU with minimal execution time ensuring the isolation property. Optimizing the transaction schedule is a combinatorial problem, which is ideal to be solved by quantum annealers as special form of quantum computers. In our contribution we show how to transform an instance of the transaction schedule problem into a formula that is accepted by quantum annealers including a proof of validity and optimality of the obtained result. Furthermore, we analyze the number of required qubits and the preprocessing time, and introduce an approach for caching formulas as result of preprocessing for the purpose of reducing the preprocessing time. In an experimental evaluation, the runtime on a quantum annealer outperforms the runtime of traditional algorithms to solve combinatorial problems like simulated annealing already for small problem sizes

K(2P)18.1 translates T cell receptor signals into thymic regulatory T cell development
It remains largely unclear how thymocytes translate relative differences in T cell receptor (TCR) signal strength into distinct developmental programs that drive the cell fate decisions towards conventional (Tconv) or regulatory T cells (Treg). Following TCR activation, intracellular calcium (Ca2+) is the most important second messenger, for which the potassium channel K(2P)18.1 is a relevant regulator. Here, we identify K(2P)18.1 as a central translator of the TCR signal into the thymus-derived Treg (tTreg) selection process. TCR signal was coupled to NF-kappa B-mediated K(2P)18.1 upregulation in tTreg progenitors. K(2P)18.1 provided the driving force for sustained Ca2+ influx that facilitated NF-kappa B- and NFAT-dependent expression of FoxP3, the master transcription factor for Treg development and function. Loss of K(2P)18.1 ion-current function induced a mild lymphoproliferative phenotype in mice, with reduced Treg numbers that led to aggravated experimental autoimmune encephalomyelitis, while a gain-of-function mutation in K(2P)18.1 resulted in increased Treg numbers in mice. Our findings in human thymus, recent thymic emigrants and multiple sclerosis patients with a dominant-negative missense K(2P)18.1 variant that is associated with poor clinical outcomes indicate that K(2P)18.1 also plays a role in human Treg development. Pharmacological modulation of K(2P)18.1 specifically modulated Treg numbers in vitro and in vivo. Finally, we identified nitroxoline as a K(2P)18.1 activator that led to rapid and reversible Treg increase in patients with urinary tract infections. Conclusively, our findings reveal how K(2P)18.1 translates TCR signals into thymic T cell fate decisions and Treg development, and provide a basis for the therapeutic utilization of Treg in several human disorders.Peer reviewe
Using gamification to transform the adoption of servitization
Increasingly, manufacturing organizations compete by developing product-service systems rather than offering products alone. To transform themselves into providers of advanced services, however, product-centric manufacturing firms need to overcome a range of barriers. While previous studies have highlighted the teaching/learning potential of 'gamification' (the use of ideas and techniques found in game-playing), the opportunities to harness this approach to help tackle such barriers have yet to be fully realized. Our study extends the debate by integrating established frameworks relating to emotional mechanics of gamification with the adoption of advanced services, arguing that such mechanics can facilitate and strengthen companies' transformation into advanced-service providers. Based on a systematic analysis of nearly 90 selected publications, we develop six conceptual propositions to explore how gamification can aid the transformation process. Our findings will help both practitioners and researchers apply emotional mechanics of gamification when seeking to address different hurdles hindering the development and provision of advanced services

The first non-human INPPS (International Nuclear Power and Propulsion System) flagship flight with orbits Earth-Mars-Earth-Jupiter/Europa (after 2025) is the most maximal space qualification test of INPPS flagship to carry out the second INPPS flagship flight to Mars with humans (in the 2030th). This high power space transportation tug is realistic because of A) the successful finalization of the European-Russian DEMOCRITOS
and MEGAHIT projects with their three concepts of space, ground and nuclear demonstrators for INPPS realization (reached in 2017), B) the successful ground based test of the Russian nuclear reactor with 1MWel plus important heat dissipation solution via droplet radiators (confirmed in 2018), C) the space qualification of the Russian reactor by 2025 and D) the perfect celestial constellation for a Earth-Mars/Phobos-Earth-Jupiter/Europa trajectory between 2026 and 2035.
Therefore the talk sketches the preparation status of INPPS flagship with its subsystems. Critical performance will be studied by parallel realizations of the ground and nuclear demonstrators of DEMOCRITOS (until 2025). The space qualification of INPPS with all subsystems including the nuclear reactor in the middle of
the 2020th plus the INPPS tests for about one to two years - first in high Earth orbit robotic assembly phase of INPPS and later extended in nearby Earth space environment flight - means a complete concepts driven approval for all applied INPPS space subsystem technologies.
It is also important to consider wider aspects for the overall mission implementation phase. Component like the nuclear reactor as the power source for the propulsion system will have to agree with the 1992 UN principles relevant to the use of nuclear power sources (NPS) in outer space. Therefore this talk will look into the legal and policy issues of nuclear space systems related to the international realization of mission design,
requirements of associated safety regulations (including AI applications in the subsystems) and new aspects for
INPPS flagship commercialization and new media communication on board
A communal catalogue reveals Earthâs multiscale microbial diversity
Our growing awareness of the microbial worldâs importance and diversity contrasts starkly with our limited understanding of its fundamental structure. Despite recent advances in DNA sequencing, a lack of standardized protocols and common analytical frameworks impedes comparisons among studies, hindering the development of global inferences about microbial life on Earth. Here we present a meta-analysis of microbial community samples collected by hundreds of researchers for the Earth Microbiome Project. Coordinated protocols and new analytical methods, particularly the use of exact sequences instead of clustered operational taxonomic units, enable bacterial and archaeal ribosomal RNA gene sequences to be followed across multiple studies and allow us to explore patterns of diversity at an unprecedented scale. The result is both a reference database giving global context to DNA sequence data and a framework for incorporating data from future studies, fostering increasingly complete characterization of Earthâs microbial diversity
